Can I only "stick" weld with DCEP as stated on the video with my Synchro
250 or can I run the rods also on DCEN?

You should be able to stick weld in all three modes with your machine: DCEP, DCEN, or AC.

I don't understand setting the "DIG" with the Synchro when stick welding??

It adjusts the OCV/amps for starting the arc. once the arc is started it reverts to your preset on the dial

In short, from my understanding, dig depends on arc length. Hot start will do as described above. Do a search for DIG and start reading. There are plenty enough posts and threads now to give a pretty good informed idea of dig.
